New streamlined #CyTOF workflow for whole blood defines a circulating immune cell signature of #COVID19 from Icahn School of Medicine at Mount Sinai. The team optimized a standardized, streamlined sample processing workflow to allow whole blood to be easily stained, stabilized, preserved and transferred to a central #masscytometry core for final processing and data acquisition The workflow leverages the Maxpar® Direct™ Immune Profiling Assay™ containing a 30‐ marker immunophenotyping panel in a single-tube dried-down format. “Pre-mixed dry antibody panels offer significant advantages over a conventional liquid antibody for #flowcytometry workflows by significantly reducing the processing time, technical variation and potential errors associated with pipetting multiple individual #antibodies, and these advantages are amplified when using larger antibody panels.”

A group at Washington University School of Medicine in St. Louis discovered that #glioblastoma #stemcell subpopulations differ in signaling, self-renewal potential, and in vivo tumorigenicity. Knowledge of marker expression among these populations could help better understand tumor heterogeneity and treatment resistance. #Masscytometry was used to evaluate the intracellular states associated with four common GSC surface markers - measuring proliferation, migration, and tumorigenesis, core developmental pathways, their downstream effectors, and cancer-associated markers and finding 15 distinct GSC subpopulations in patients that correlated with outcome. “Mass cytometry… largely overcomes the spectral overlap typical of standard flow cytometry, which limits the number of observations possible on a given cell,” the paper states. https://lnkd.in/g6V-qZw

Diagnosis of a pulmonary embolism can be challenging due to a variable and often non-specific clinical presentation. It is a complex multi-factorial disease caused by external and multiple genetic factors, but studies tend to focus on risk factors associated with venous thromboembolism. We would like to highlight this interesting work using #microfluidics. A team at Hannover Medical School conducted a large case-control genetic association study that evaluated the association of SNPs that are robustly associated with venous thromboembolism or deep vein thrombosis in previous studies in subjects who died from pulmonary embolism.
